The current study reports an integrated microfluidic system for rapid serological analysis of antibodies associated with the infection of dengue virus by utilizing virus-holding magnetic beads. This microfluidic system integrates micropumps, a four-membrane-type micromixer, and on-chip microcoils array to perform the rapid detection of immunoglobulin G (IgG) and immunoglobulin M (IgM) simultaneously. Using virus-holding magnetic beads, IgG and IgM in a clinical serum samples can be captured, followed by purification of these beads utilizing the on-chip microcoils array. Finally, the optical signals of sandwich-like magnetic complexes can be detected by an optical detection module. The whole reaction time is measured to be less than 30 minutes. More importantly, the detection limit has been improved to 21 pg. ©2009 IEEE.
