We developed a method for uniform and high-density immobilization of antibodies in nanochannels. A peptide reagent that specifically and covalently binds to the Fc region of the antibody was introduced to control the orientation of the antibody. Compared with the conventional immobilization method using glutaraldehyde, the density of antigen-adsorbed sites was 12 times higher and the coefficient of variation of density was one-sixth. We observed that antigens introduced into the nanofluidic channel were rapidly captured within 0.5 seconds.
