This paper reports mitochondrial DNA damage analysis in single cells using a microfluidic platform. DNA damage and single-nucleotide polymorphisms (SNPs) in individual mitochondrial DNA molecules were analyzed using padlock probes, made visible by rolling-circle amplification of the circularized padlock probes. By combining microfluidcs with padlock probes, individual cells have been manipulated and investigated with single-molecule sensitivity.
