Abstract
A liquid chromatography using 100-nm scale nanofluidic channel was designed and developed to realize one million theoretical plate numbers. Higher separation efficiency and smaller detection volume have been important goals for all separation analyses, although conventional liquid chromatography using packed column has a limitation of the performances. Contrary, we have developed a chromatography using nanochannel as a separation column and enabled ultrahigh separation efficiency breaking the limitation of conventional chromatography. However, in order to realize high separation performance, longer nanochannel and higher pressure are necessary. In this study, theoretical plate number was designed and ultrahigh pressure system was developed for one million plates. In addition, nanochannel was newly designed to exploit higher pressure and verified to discuss a feasibility of million plate numbers.
