Abstract

We developed ELISA device utilizing thin-layered channels, which have millimeter-scale width and nanometer-scale depth. High surface-to-volume ratio (S/V) of thin-layer channels enabled 100% capture of target molecules. By investigating conditions to achieve 100% capture, coefficient of variation (C.V.) of ELISA signal was reduced. Target molecules in 1 PL sample were accumulated in a 20 nL reaction chamber. Here, the limit of detection (LoD) of C-reactive protein was reduced from 17 pM to 0.77 pM.
