This paper reports analysis of DNA damage, both in mitochondrial DNA (mtDNA) and nuclear DNA in single cells using a microfluidic platform and padlock probes. Damage in DNA such as strand breaks were analyzed in situ using padlock probes, made visible by rolling-circle amplification (RCA) of the circularized padlock probes [1]. The correlation between DNA damage / damage repair and single nucleotide variation in individual mitochondrial DNA also have been studied. © 2008 CBMS.
