Abstract

Two novel microfluidic devices, one silicon (Si)-and the other poly(dimethylsiloxane) (PDMS)-based, that incorporate arrays of nanoholes were designed and fabricated for nanofluidic interrogation of large number of single cells simultaneously. The electrical properties of the devices were characterized, which compared very well with those of traditional micropipettes. Cells cultured in finished devices appeared morphologically normal and viable. The devices are suitable for patch-clamp electrophysiological recordings of ion channel activity [l] as well as for the focal delivery of biochemical factors to the cell surface with submicron-scale precision.NANOHOLE ARRAYS FOR PARALLEL PATCH- CLAMPING AND FOCAL DELIVERY OF BIOCHEMICAL FACTORS TO CELLS (PDF Download Available).
