Proteins at the biomaterial interface include surface-adsorbed proteins and soluble proteins secreted by cells adherent or attached to biomaterial interfaces. These proteins carry multiple functions such as directing cell adhesion, proliferation, differentiation, and migration. The identification of proteins at the material interface will help elucidate the impact of proteins in mediating cell-material interaction, host response, and cell signaling pathway. The aim of this chapter is to provide an overview of the application of proteomic tools to analyze proteins at the material interface. Two main analytical methods, protein microarrays and mass spectrometry (MS), have been reviewed and their applications in proteomic study of biomaterial-related proteins were discussed in case studies. In sum, proteomics provides a viable approach to survey global proteome of surface-adsorbed proteins and soluble proteins from adherent cells at the biomaterial interface. © 2012 American Chemical Society.
