Abstract
Single molecule detection and manipulation are the most intriguing topics in nanotechnology in these ten years, and the most useful single molecule approaches are optical tweezers and single particle tracking. The position detection methods with nanometer resolution are very important in single molecule studies. For this purpose, we use LabVIEW-based two-dimensional fast Fourier transform (2D FFT) software for cross-correlation process which can be used to determine the object displacement between two consecutive images with nanometer resolution. Using cross-correlation analysis, we calibrate a two-trap optical tweezers system and measure the fluid temperature in microdomains. Finally, cell motility and single vesicle behavior are revealed by analyzing the consecutive bright-field cell images.
