Abstract
Plants are frequently exposed to low temperature stress both in natural and agricultural conditions, which often result in great damage to plant yield and to people demand. Thus, understanding the physiological processes that underline stress injury and adaptation and acclimation mechanisms of plants to environment stress is of immense importance to both agriculture and the environment. PCR-select cDNA subtraction was performed to isolate the cold inducible genes in mungbean (Vigna radiata) VC1973A cultivar (chilling-sensitive). A total of 263 ESTs was obtained and combined to other mungbean ESTs in our laboratory. To sum a total of 1,022 ESTs and all subjected to cDNA Microarray analysis. In order to understand the role of polyamines and ethylene between cold-sensitive and cold-tolerant plant during cold acclimation, we applied Northern blot and RT-PCR to investigate the temporal pattern of VrSAMDC (S-adenosylmethionine decarboxylase), VrSAMS (S-adenosylmethionine synthetase), and VrACO (1-aminocyclopropane-1-carboxylic acid oxidase) transcripts during exposure to low temperature both in VC1973A cultivar (cold-sensitive) and NM94 cultivar (cold-tolerant).
