AbstractIn order to obtain the higher resolution images from confocal microscope, a method has been developed to construct the whole 3D image based on several sub-image stacks. High-resolution images were obtained from scanning frontal and rear side of samples at specific area several times. The images were processed by image registration via XY plane and then Z-axis Montage. The XY plane montage is to combine the two individual 2D images from two sub-image stacks. The cross-correlation method was used to find the translation of X and Y axises, meanwhile decreased resolution of image was adopted to speed the computer calculation. The Z-axis montage is to assemble two 3D image stacks in Z-axis direction. The fast Fourier transform (FFT) was used to check if the frontal and rear side of scanning stacks overlap or not. If there is overlap, using FFT, Sobel edge detector and cross-correlation determine the overlap position. Then using the images of the overlap layer compute the translation of XY plane and rotation angle (θ) of Z-axis by FFT. After processing, adjust the position of the frontal and rear view of scanning stacks to obtain the whole 3D images.In order to demonstrate this program, we use biological brain images from fruit fly, D. melanogaster. The results show that the 3D image stacks were successfully combined into the intact and high-resolution 3D image.
