Abstract
The most common tool for taking 3D images is a confocal laser scanning microscopy (CLSM). By using CLSM, which generates optical slices, people can get the slices of a bio-sample without cutting or damaging it. After reconstructing the optical slices, we can get a three-dimension image of the sample. However, there is still a weakness of CLSM. That is the Z axial resolution. As we know, CLSM has high resolution in X-Y plane and the resolution is much higher than the resolution in Z axial direction. To improve the resolution in Z axial direction, this research is demonstrating a “Novel Confocal Microscopy System with Revolvable stage” which can be used in the observation of fly brain’s neuron maps. There are three grooves at the metal sample holder. Each groove will be at a suitable size for the fly brain, whose size is about 0.5 x 0.25 x 0.15 mm. After anchoring the fly brain into the grooves, we will be able to rotate the metal sample holder to take the images of the fly brain from all directions. Furthermore, this research use the technique of image fusion, which is including deconvolution, registration and interpolation, to reconstruct all the images and form a high resolution 3D image, whose resolution of Z direction is improved.
