Abstract

Since 1995 the Patrick O. Brown’ lab in Stanford University in USA developed the system of cDNA microarray, the cDNA microarray have instantly stirred up a new trend in biological and medical research. This system is a largescale gene expression investigation tool, including an arrayer system and a scanner system. The cDNA microarray, one type of gene chip systems, can study 6000–10000 genes’ expression simultaneously; therefore, the cDNA microarray is a quite powerful technology in studying gene expression.The studies of this thesis include two parts: one is the study of the response of MRC-5 cells to UV irradiation, and the other is the optimization of cDNA Microarray. In order to develop a system for UV irradiation, we firtstly studied the UV irradiation system to find out a stable and reliable condition for experiment. Further, by the use of MTT assay and flow cytometry experiments, we decided the exposure dose and the observing time. On the optimization study, the mRNA amplification, fluorescent dye incorporation, and the hybridization using RNA as the target were proceeded. Here the thesis employed in vitro transcription to amplify the mRNA to get a linear and miss less amplification. Unlike the conventional way, we utilitied the combination of aminoallyl and NHS-ester to incorporate the fluorescence molecular to the RNA. Finally, we use the labeled aRNA instead of cDNA to hybridize with probe. The goal is to improve the signal and the S/N ratio, and make the result be more dependable.The thesis is just a preliminary study, and the result is not very complete. But the approach could be affirmed. We hope the research could be a guide for the following research, and be useful in the clinical study about the UV damage.
