Abstract

甘草 甘草酸 豬苓 克速鎮 實驗性輻射損傷 脾臟 白血球 去氧核糖核酸 細胞免疫力 γ-射線 Glycyrriza glycyrrhizic acid Polyporus umbellatus cellular immunocompetence γ-irradiation
本研究論文分成四部分,分別說明於下:第一部分旨在探討γ-射線對於鼷鼠細胞免疫力之效應。本研究包含Ⅰ-A及Ⅰ-B兩部分:Ⅰ-A部分在探討5、50及100 cGy的γ-射線對鼷鼠白血球及其分類計數,以及脾臟之損傷。結果顯示:在100 cGy照射後,鼷鼠之白血球及其分類計數、脾重及相對脾重顯著下降。50 cGy照射後,僅見白血球及其分類計數下降。然而,5 cGy的照射,皆無明顯抑制作用。Ⅰ-B部分在探討100、200、300及400 cGy的γ-射線對鼷鼠細胞免疫力之效應。結果顯示:在分別照射1至4 Gy (葛雷)的γ-射線後,各組鼷鼠之白血球及其分類計數皆見下降,脾臟細胞對細胞增殖素如PHA、PWM、Con A及LPS刺激後之變形反應亦見下降,且隨著劑量增加,而加大其抑制程度。顯示1至4 Gy的γ-射線對鼷鼠細胞免疫力具有顯著的抑制作用。第二部分旨在探討甘草及甘草酸對於照射過 4 Gy的γ-射線之鼷鼠細胞免疫力之效應。結果顯示:甘草及甘草酸對於被γ-射線所抑制之鼷鼠白血球及其分類計數,以及脾臟細胞對PHA、PWM、Con A及LPS之變形反應,均有促進提早恢復之作用。顯示甘草及甘草酸對經γ-射線抑制的鼷鼠,可促進恢復其細胞免疫力。第三部分旨在探討甘草與甘草酸對於照射過4 Gy的γ-射線鼷鼠脾臟損傷之修復作用。結果顯示甘草及甘草酸對於照過γ-射線鼷鼠之脾臟損傷具有顯著的促進修復作用。第四部分旨在探討豬苓及速克鎮對於照射過 1 Gy的γ-射線之鼷鼠細胞免疫力之效應。結果顯示:豬苓及克速鎮對於被γ-射線所抑制之鼷鼠白血球及其分類計數,以及脾臟細胞對PHA、PWM、Con A及LPS之變形反應,有促進提早恢復之作用。顯示豬苓及克速鎮對經γ-射線抑制之鼷鼠,可促進其細胞免疫力之恢復。The study was divided into four parts described as in thefollowing: Part Ⅰ. The effects of 137Csγ-ray irradiation (RT)on cellular immunocompentence (CI) of mice were investigated inthis part. The dosage of RT, including 5, 50, 100, 200, 300and 400 cGy. The results revealed that all dosage of 100, 200,300 and 400 cGy of RT played an evident inhibitory effects oncounts of leukocyte (Leu) and its differential counts (DC), thespleen weight (SW) and the blastogenic response of splenocytes(BRS) to mitogens, such as PHA, PWM, Con A and LPS in mice.The higher the dosage ofγ-irradiation, the more inhibition ofCI in mice was found. Prat Ⅱ. The effects of Glycyrrizae radix(GL) and Glycyrrhizic acid (GA) on CI in 4 Gy γ-ray irradiatedmice was investigated in this part. The results revealed thatGA and GL were effective in enhancing the recovery of leukocyteand its differential counts, and the BRS to PHA, PWM, Con A andLPS from the inhibition of 4 Gy γ-irradiation. GA and GLcould help the recovery of the CI in the γ-ray irradiatedmice. Part Ⅲ. The effects of GA and GL on promoting therecovery of mice from radiation injury was investigated in thispart. The results revealed that GA and GL had the beneficialeffects to restoration of radiation injury in spleen of mice.Part Ⅳ. The effects of Polyporus (PU) and PSK (Krestin) on CIof 1 Gy γ-ray irradiated mice was investigated in this part.The results revealed that PU and PSK were effective inenhancing the recovery of Leu and DC, and the BRS to mitogensfrom the inhibition of 1 Gy γ-irradiation. PU and PSK couldhelp the recovery of the CI in the γ-ray irradiated mice.
