When scanning biological samples with atomic force microscopy(AFM), we face some difficulties due to soft and mobile properties of the specimens. Furthermore,there are interferences from scan environments in liquid state. In this thesis, we mainly focused on biological sample preparation for imaging with AFM. The image scanning and force curve measurements were discussed. In the subject of adherent cells sample preparation, we chose SiHa, the human cervical cancer epidermal-like cell lines for instance. First, we constructed microstructure arrays by photolithography, and then coculture cells and arrays. Before AFM scanning, cell fixation processes were performed. We discussed the interactions between cells and microstructure arrays, which can be used to improve the procedures of sample preparation. For suspended cells, we used erythrocytes which were purified and concentrated through the human venous blood centrifugation. Cell geometry, morphology imaging, and cell elasticity were measured by AFM.
