Abstract
There were a lot of analyzing or training systems developed to study behavior of fruit flies, Drosophila melanogaster in the past. In order to observe the different behaviors, these systems have their customized set of tools or special functions. However, there was no team that has successfully developed the real-time analysis with outside stimulated system which can do the Chlamydomonas reinhardtii channelrhodopsin (ChR2) and Natronomonas pharaonis halorhodopsin (NpHR) experiments. In this thesis, we present a “Camera-Based Real-Time Analysis with Two Lasers Stimulated for Behavior Study of Drosophila”. The system use image processing to get the fly’s position and recognize the two different flies. After analysis, it will rotate the two-axis motor to make two laser stimulations track the specific fly. Though this way, system can do the experiments of optogenetics and know the relationship between behaviors and optogenetics. This System is based on the 1st. system “Camera-Based Real-Time Analysis with Laser Stimulated for Behavior Study of Drosophila” which is developed by our laboratory. The new version not only can do the all things which the previous system can do, but also can track the more detailed positions (head, thorax and abdomen) and provide more flexibility for experiments by controlling two shutters. Besides, the new version also improves the shaking problem in the previous system.
