Abstract
本研究旨在比較篦麻油乳化的紫杉醇 (CET) 與類脂小體包裝的紫杉醇 (LET) 的抗癌效應,並探討CET或LET與X射線及一些中藥在治療實驗性肝癌的合併效應。研究結果顯示:腫瘤對照組在60天內死亡率(MST60)為 19.4天。以各種劑量之 CET 單獨治療時,最大的 MST60 (35.5天)是出現在90 mg/kg 的CET3處理組;而以 LET 單獨處理時,最大的MST60 (41.9天) 則是以120 mg/kg 的 LET4 處理組。在各種合併治療的研究中發現,療效最佳的處理方式是以90 mg/kg的 LET 與16 Gy 的X 射線之合併治療,其MR60與 MST60分別為 73.3% 和43.2天。用流式細胞術測定細胞週期的結果顯示:無論 CET 或 LET 都會造成 G2/M期的細胞增加,且隨著紫杉醇劑量的增加,就會有愈多的細胞被阻擋在G2/M期,而LET 造成的 G2/M 阻擋的程度會較 CET 處理者來的高,這個現象可由在生物分佈實驗中所得到的結果來解釋,也就是由於 LET 在腫瘤內的分佈濃度要較 CET 處理者為高。然而,部分被阻擋的細胞會從 G2/M 期逃脫掉,而形成多微核細胞,這些微核細胞最後還是會注定走向死亡。In this research, we compared the antitumor effects betweencremophor EL-emulsified taxol (CET) and liposome-encapsulated taxol (LET). The combined effects of CET, LET,X-ray and some Chinese herbs on experimental liver cancer werealso investigat- ed. The results revealed that the mortalityrate (MR60) was 19.4 days. In the treatment of CET alone,the maximun MST60 (35.3 days) was found in 90 mg/kg treatedgroup. In the treatment of LET alone, the maximun MST60(41.9 days) was found in 120 mg/kg treated group. The besttherapeutic effect was found in the group treated with 90 mg/kg of LET3 and 16 Gy of X-ray irradia- tion, its MR60 was73.3 % and 43.2 days respectively. The re- sults ofanalysis with flow cytometr showed that CET and LET couldincrease the cell population in G2/M phaseof the cellarrested in G2/M phase. The level of G2/M arrested by LETwas higher than that treated with CET. It may be explainedby the results of the CET and LET. That is, theconcertration of the biodistribution of CET and LET. Thatis, the concentration of LET localized in tumor was higherthan that treated with CET. Some of th e arresred cellsperhaps escaped from G2/M phase and formed manymultimicronucleated cells. However, they were doomed to dieeventually.
