Abstract
Enterovirus 71(EV71), belonged to picornaviriade enterovirus, caused a serious outbreak in Taiwan in 1998. In this research, we investigated high cell density culture in microcarrier culture system to produce EV71 for inactive vaccine production process. In our results, EV71 is more stable when culture in 32℃ than in 37℃. When MOI=1, Vero cell produced higher titer of EV71 than WI-38 and MRC-5. We also found out higher titer of EV71 can produce when MOI=1. In microcarrier culture system, the highest cell density reach to 8.94x106 cells/ml with 10g/L Cytodex 1. And the maximal titer, 7.17x107 TCID50/ml, also produce by Vero cells with 10g/L Cytodex 1. In 2L bioreactor, the maximal EV71 yield reach to 1.71x107 TCID50/ml by Vero cells with 5g/L Cytodex 1. The highest cell density, 1.68x106 cells/ml, was reached by 10g/L Cytodex 1 culture Vero cell in bioreactor. In this paper, we investigated several important culture factors in animal cell culture to produce EV71and We hope our results can improve the EV71 vaccine production.
