Abstract

Baculovirus/insect cell system has been widely used for recombinant protein production, but traditional system eventually resulted in cell lysis, so that the expressed recombinant protein was lost into medium. To improve this, the thesis researched the non-lytic baculovirus expression system by two different ways. First, Dr. Yu-Chan Chao at the Institute of Molecular Biology, Academia Sinica, constructed a recombinant baculovirus. In the adherent culture, the virus did not result in cell lysis after the insect cells were infected. In the thesis, we tried to transfer the novel non-lytic system to suspension culture in spinner flask. Second, some reports utilized the baculovirus as a gene delivery vehicle into mammalian cells and did not have any bad effect on the growth of mammalian cell. So, the thesis tried to develop the non-lytic mammalian/baculovirus expression system by investigated the parameters influencing the level and the duration of the expression, such as medium exchange, superinfection and virus transduction strategies.
