本發明提供一種醫藥組合物,其包含膜衣層以及藥物層。所述藥物層被膜衣層包覆,且藥物層包含有效劑量的疏水性小分子藥物、有效劑量的親水性小分子藥物、親脂性溶劑、酸性化合物和泡騰成分。當所述醫藥組合物經口服進入腸道時會產生二氧化碳氣泡,藉由二氧化碳氣泡膨脹、破裂引起的擾動,使醫藥組合物經自我乳化形成帶有疏水性小分子藥物的脂質油滴,並使親水性小分子藥物溶解於腸液中。藉此,所述醫藥組合物不需使用帶有毒性之載體,可經由口服同時傳遞疏水性小分子藥物和親水性小分子藥物,並可增加藥物的溶解度以提高口服生物利用度。
