Abstract

The invention provides an escherichia coli gene editing system. The escherichia coli gene editing system comprises escherichia coli, helper plasmids and donor plasmids. Wherein the helper plasmid comprises a transposase compound expression cassette, a Cas12k expression cassette, a first sgRNA cassette, a first antibiotic resistance gene and a first replication starting point, and the donor plasmid comprises a ShCAST transposon left-end sequence, an exogenous gene expression cassette, a ShCAST transposon right-end sequence, a second sgRNA cassette, a second antibiotic resistance gene and a second replication starting point. According to the Escherichia coli gene editing system, a large-fragment DNA sequence can be simply, conveniently and effectively embedded into a target chromosome, and double-strand breakage of an Escherichia coli genome cannot be caused, so that useful Escherichia coli strain metabolic pathways in different biotechnologies can be adjusted.
