Abstract
The invention provides a gene editing system of Candida vesii. The gene editing system comprises Candida vesii, a first gene editing segment and a second gene editing segment. The first gene editing segment includes a first homologous arm and a screening gene. The second gene editing segment comprises a second homologous arm, a Cas9 expression box and an sgRNA box. The Cas9 expression cassette comprises a Cas9 promoter, a Cas9 gene and a triple-core localization sequence. The sgRNA box comprises a sgRNA promoter, a first ribozyme, a targeting sequence, a scaffold sequence and a second ribozyme. The first gene editing segment and the second gene editing segment form a linear segment for performing gene editing on the chromosome of the Candida vesii. Therefore, each set of target chromosome of the candida vesii can be edited conveniently, and a stable transformant can be obtained without any specific condition.
